Chemical Constituents and Antipyretic Effect of Eupatorium lindleyanum DC.

Abstract: In this study,chemical composition of the ethanol extract of Eupatorium lindleyanum DC.was analyzed by UPLC/Q-TOF-MS.Six compounds were isolated from E.lindleyanum. These compounds were identified as vanillic acid (1),n-octacosanoic acid (2),β-sitosterol (3),daucosterol (4),linarin (5),5,8,4′-trihydroxy-7,3′-dimethoxy flavone (6). Compound 2,5 and 6 were obtained from this plant for the first time.In addition,the antipyretic effect and mechanism of E.lindleyanum on LPS-induced febrile response in rats was observed and elucidated.After the LPS challenge,the temperature of rats was recorded every 30 min,it lasted for 6 h,and then 7 h,8 h.Myeloperoxidase (MPO) activities in liver tissues was examined,the levels of inflammatory cytokine tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in serum were measured using ELISA.As a result,the rats of model group showed obvious three-phase fever and the heating peak value (1.97 ℃) appeared at 4 h.The ethanol extract of E.lindleyanum (EUP-EtOH) significantly decreased the body temperature of rats,and at 8 h following LPS challenge effectively inhibited MPO activity in liver tissues,TNF-α and IL-6 levels in serum.
