Accelerated Solvent Extraction and DPPH Free Radical Scavenging Activity of Triterpenoids from Ganoderma flexipes

Abstract: In this study,accelerated solvent extraction (ASE) method was used to extract triterpenoids from Ganoderma flexipes. Single factor and L9(33) orthogonal experiments was applied to optimize the extraction conditions.In addition,free radical scavenging activities of the extracts were evaluated by DPPH assays.The results showed that the optimal extraction conditions were 60% methanol (chloroform and methanol) as the extraction solvent,extraction temperature of 180 ℃,extraction time of 12 min and with 2 times of static extraction,in which extraction temperature had extremely significant effect on the extraction yield of triterpenoids.Under the optimized conditions,the extraction yield of total triterpenoids from G.flexipes was up to (2.14±0.18)%.In the DPPH experiments,the IC50 value of total triterpenoids obtained from the optimal ASE extraction process and the traditional methods were 129.5 and 273.4 μg/mL,respectively.Comparing with the traditional heat reflux extraction,ASE was simple,rapid,economical,practical,which can be used for the extraction of triterpenoids from G.flexipes.
