Protective Effect and Mechanism of Fisetin on Aβ42-induced Rat Model of Alzheimer's Disease

Abstract: Rats were administered bilaterally in hippocampal CA1 area by injecting Aβ42 to produce an animal model of AD,where animal behaviors were detected by the passive-avoidance test.The contents of GSH,MDA,and PC in brain tissues were extensively evaluated to investigate the effect of fisetin on antioxidant activity and memory function.In addition,the spleen index,thymus index,liver index and the expression of GFAP were used to preliminarily assess the anti-inflammatory and anti-aging effect of fisetin on AD model rats.The results showed that the memory function was significantly reduced in model group of rats,and fisetin can improve memory impairment of AD model rats.Moreover,compared with the model group,the spleen index,thymus index,liver index and the content of GSH in brain tissues were significantly enhanced whereas the expression of GFAP,MDA and PC contents were decreased,especially in the group that received 100 mg/kg fisetin.Thus fisetin might be a potential agent for treatment of Alzheimer’s disease via inhibiting oxidative damage and activation and proliferation of astrocyte reducing and further reducing nerve inflammation.
