Effects of Propolis Flavonoids Pinobanksin-3-acetate on the Expression of HSPA6,GEM,FOS,RGS2and GADD45G Gene in Human Gastric Cancer Cell Line SGC-7901

Abstract: In order to investigate the effects of Propolis Flavonoids Pinobanksin-3-acetate(PB3A) on gene expression changes in human gastric cancer cell line SGC7901,the cellline was treatedwith different concentrations ofPB3A(0,40,80 μg/mL),and themRNAexpressionof HSPA6,GEM,FOS,RGS2 and GADD45G geneswas detected by real-time quantitative PCR.The result showed that after treated the cell line with 40 μg/mL of PB3A,the expression of HSPA6, GEM and FOS genes were significantly increased (P<0.05).The differential expression rates were 0.12(HSPA6),2.17 (GEM) and 2.1 (FOS) respectively.However,theexpressionof GADD45G and RGS2 were not significantly increased(P>0.05)incomparison with untreated control group;After treated the cell linewith 80 μg/mL of PB3A,the expression of HSPA6 gene was significantly (P<0.05) increased,the differential expression rate of HSPA6 gene was 0.5,and the expressionof GEM, FOS, RGS2 and GADD45G geneswere not significantlyincreased (P>0.05) by comparing to the control group.In conclusion,It was deduced that PB3A affected gastric cancer cell proliferation through regulating the expression of HSPA6, GEM and FOSgenes.
