Effects of Dendrobium candidum Polysaccharide on Immunity,Intestinal Microbiota and Enzyme Activity in Mice with Spleen Deficiency Constipation

Abstract: To explore the effects of Dendrobium candidum polysaccharide on immunization,intestinal microbes and enzyme activities in mice with spleen deficiency constipation,and clear the role of polysaccharide in the treatment of spleen deficiency constipation.The mouse spleen deficiency constipation model was established by giving senna water decoction for 7 days,and then restricting their diet to make them hunger satiety disordered for 8 days.After the success of modeling,normal group and model group mice were treated with sterilized water by intragastric administration,the treatment groups were fed with D.candidum polysaccharide solution.Several indicators were analyzed including body weight,spleen coefficient,thymus coefficient,intestinal flora and enzyme activity.The results showed that there were significant differences in body weight,changing rate of body weight,spleen index,thymus index and the quantities of five culturable microorganisms (bacteria, Escherichia coli, Lactobacillus, Bifidobacterrium and Fungi) in the model group compared with these of normal group.After treatment,the body weight and changing rate of body weight increased significantly,higher than these of normal group;the spleen index and thymus index recovered to different degrees,closed to these of normal group; Escherichia coli recovered a lot,but far from the normal level; Lactobacillus and Bifidobacterium were significantly decreased,but the total number was significantly higher than the normal group;xylanase and protease were close to normal group,amylase was significantly increased,but cellulase significantly reduced.The results showed that D.candidum polysaccharides can promote body immunity,regulate the balance of intestinal microbiota and intestinal enzyme activity,optimize the intestinal environment and improve the symptoms of spleen constipation.
