Study on the apoptosis mechanism of human melanoma A375 cells induced by Neochamaejasmin A

Abstract: To explore the effect of Neochamaejasmin A on the proliferation and apoptosis in human melanoma cell line A375 cells.MTT (thiazolyl blue) was applied to detect the cell viability of A375 cells treatment with Neochamaejasmin A.Hoechst 33258 was employed in observing the cell morphology.Flow cytometry was applied to assess the apoptosis rate and mitochondrial membrane potential.The protein expression levels were assessed with Western Blot.Data showed that Neochamaejasmin A significantly suppressed the proliferation and growth of A375 cells at a time-concentration-dependent manner.The A375 cells were treated with differernt concentration Neochamaejasmin A (0,15,30 and 45 μmol/L) for 48 h,the cells showed significant apoptosis characteristics.The apoptosis rate was significantly increased (P<0.01) and mitochondrial membrane potential was significantly reduced (P<0.01).Western blot showed that Neochamaejasmin A upregulated the protein levels (Bax,caspase-3 and Cytochrome C) and downregulated expression level of Bcl-2 protein.The above results indicated that Neochamaejasmin A inhibited proliferation and induced apoptosis in A375 cells.Its mechanism might be related to the induction of apoptosis in the mitochondrial pathway.
