Study on the chemical constituents of the aerial parts of Polygonatum sibiricum and its pancreatic lipase inhibitory activity

Abstract:

To study the chemical constituents of the aerial parts parts of Polygonatum sibiricum and its pancreatic lipase inhibitory activity.Compounds were isolated from the ethanol extract of the aerial parts of P. sibiricum by various chromatographic techniques including Sephadex LH-20,Diaion HP20ss,Toyopearl HW-40F,ODS C18 and medium-low pressure semi-preparative liquid phase.The inhibitor activity was detected by porcine pancreatic lipase inhibitor activity screening model.As a result,a total of 14 compounds were isolated and purified.Their structures were elucidated on the basis of NMR,MS spectroscopic and comparison with literature data as methyl caffeate (1),chlorogenic acid (2),isovitexin (3),isoorientin (4),orientin (5),orientin 2″-O-xyloside (6),luteolin 6-C-β-D-galactosyl (1→2)-β-D-glucoside (7),apigenin 6-C-β-D-galactosyl (1→2)-β-D-glucoside (8),luteolin 6-C-β-D-xylosyl (1→2)-β-D-glucoside (9),orientin-6-C-β-D-glucoside (10),isovitexin 8-C-β-D-glucoside (11),chrysoeriol (12),quercetin 3-O-α-L-rhamnoside (13),n-heneicosanoic acid (14).Among them,compounds 6-14 were reported from P. sibiricum for the first time.Compounds 2,3,6,7,8,10,11,13 were screened for pancreatic lipase inhibitory activity,and the results showed that the eight compounds have certain inhibitory activity on pancrelipase.
