Study on the sedative and hypnotic active components of Diaphragma Juglandis Fructus

Abstract:

Explore the sedative and hypnotic components of Diaphragma Juglandis Fructus(DJF),and provide a theoretical basis for the application of DJF in sedative and hypnotic health products or medicines.The active constituents were extracted by ethanol and water,separated and purified by multiple chromatography,and then identified the chemical structure by NMR.The results showed that the water eluate separated by the macroporous resin from the extract of DJF had a significant sedative and hypnotic effect,and then separated by MCI,ODS and other columns.From this part,four monomer compounds are obtained,respectively 2-(hydroxymethyl) oxane-3,4,5-triol (1),3-hydroxy-4-iminobutanoic acid (2),(1′-methyl-2′-hydroxy) propane-O-α-D-glucopyranoside (3),6-hydroxy naphthyl-1,2-(2-O-β-D-pyran glucoside) (4).Compound 1 could only significantly prolong the sleep time of mice,while compound 2 could significantly reduce the mouse's movement speed and time,increase the time spent in the central area,and could shorten the incubation period and prolong the sleep duration.DJF is a natural active product with potential sedative and hypnotic,and its research and application in the field of health care has important research significance and development value.
