Study on the effect and mechanism of resveratrol on rats in renal fibrosis with high-intensity exercise

Abstract:

This study investigated the effect and mechanism of resveratrol on rats in renal fibrosis with high-intensity exercise.Male SD rats were randomly divided into control group (CONT),resveratrol group (RESV),exercise-induced kidney injury model group (EIKIM),and resveratrol-intervened exercise-induced kidney injury model group (REIKIM).Specimens of blood and renal tissues were harvested 24 h after the final training to evaluate the histomorphology and extent of fibrosis in the rat kidneys,in addition to examining pertinent biochemical parameters.The findings revealed that,compared to CONT group,EIKIM group exhibited significant pathological changes and fibrosis in renal tissues.Serum creatinine (Cr) and urea nitrogen (UN),as well as renal collagen deposition scores,protein expression levels of phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK),nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB),acetylated nuclear factor kappa-light-chain-enhancer of activated B cells (Ac-NF-κB),transforming growth factor beta-1 (TGF-β1),tumor necrosis factor alpha (TNF-α),and interleukin-1 beta (IL-1β) levels were markedly increased (P<0.01).Conversely,the protein expression level of silent information regulator 1 (SIRT1) was significantly decreased (P<0.01).Compared to EIKIM group,REIKIM group demonstrated a marked improvement in pathological changes and fibrosis in renal tissue.Notably,serum levels of Cr and UN,as well as renal collagen deposition scores,protein expression of p-p38 MAPK,NF-κB,Ac-NF-κB,and TGF-β1,TNF-α,IL-1β levels were significantly reduced (P<0.05,P<0.01).Conversely,the SIRT1 expression was significantly elevated (P<0.01).In summary,resveratrol effectively suppressed the renal inflammatory response induced by high-intensity exercise in rats by modulating the expression of key proteins in p38 MAPK/SIRT1/NF-κB signaling pathway,thereby mitigating the progression of renal fibrosis and preserving renal structure and function.
