Purification of Earthworm Protein from Coelomic Fluid and Its Bioactivity

Abstract:

The coelomic fluid of earthworm Eisenia foetida exhibit extensively biological activity previously. Earthworm was induced by 6-Voltages electric stimulation to secrete coelomic fluid, followed by centrifugation and ultrafiltration to obtain a crude protein mixture. The crude extracts have been purified by means of gel chromatography and anion-exchange chromatography in sequence, as resulted in obtaining a kind of pure protein. Ultraviolet analysis indicated the protein absorption wavelength 280 nm, and SDS-PAGE determined the protein molecular weight 38.6 KDa. The further bioactive study showed that the protein displayed the inhibition effects on the growth of Escherichia coli in 1 h and Staphylococcus aureus in 3 h, as well as lysis effects on chicken red cells (minimum effect concentration 0.39 µg/mL). It suggested that activity of the protein could be come from by a means of damage-cell structure.
