Inhibitory Activity of Ginsenoside Rb3 on Pancreatic Lipase

Abstract:

Ginsenoside Rb3 of American ginseng leaves was isolated by the methods of silica column chromatographic and JTY resin, tested by thin-layer chromatography (TLC), and examined by high performance liquid chromatography (HPLC).The experiments studied the effects of different concentration ginsenoside Rb3 isolated from American ginseng leaves on pancreatic lipase activity, which could reflect its anti-obesity function. The effect of Ginsenoside Rb3 on porcine pancreatic lipase activity was determined by measuring released free fatty acids after incubating in substrate emulsions containing bile salts and phosphatidylcholine in vitro. Ginsenoside Rb3 inhibited the pancreatic lipase activity in dose-dependent manner at the concentrations of 0.25-1 mg/mL. It inhibited hydrolysis of about 85.11±0.409% of trioein at about 1 mg/mL of ginsenoside Rb3, which was much more higher than the American ginseng leaves extract. The article manifested that ginsenoside Rb3 has the anti-obesity function through the pancreatic lipase experiment, which inhibitory mechanism pending further studies.
