Effects of Conjugated Linoleic Acid Isomers on Gene Expression of Proteins Involved in Lipid Metabolism

Abstract: RT-PCR was employed to determine isomer-specific effects of two conjugated linoleic acid (CLA) isomers on enzymes involved in lipid metabolism in cultured 3T3-L1 adipocytes.Concentration of either isomer was 75.4 μmol/L and treatment period was 8 days.mRNA levels of proteins involved in energy expenditure,apoptosis,fatty acid oxidation and lipolysis were identified.The results showed that gene expression of UCP1,UCP3,Perilipin and PPAR? increased whereas UCP2 (P<0.01) reduced for both CLA isomers.And compared with treatment of cis-9,trans-11 CLA,trans-10,cis-12 CLA significantly up-regulated mRNA levels of PKA (P<0.05),CPT-1 and TNF-α (P<0.01).Compared with control,both isomers did not significantly influence gene expression of HSL,ATGL,ACO and Leptin.
