Effect of Extraction Process and Plant Growth Regulators on Flavonoids Content in Callus of Apocynum venetum

Abstract: Optimization of flavonoids extraction process carried out in callus of Apocynum venetum through the response surface method after single factor was studied. The method indicated that there were significant effects to flavonoids content in extracting reagent and temperature. The optimal parameter for extracting flavonoids were conformed by analysis of response surface method as follows: the extracting reagent is 70% methanol, the ratio of solid to liquid is 1:40, the extracting time was 4 h, the extracting temperature is 70 ℃. Flavonoids were measured respectively from the callus induced for 100 d on 30 kinds of different concentration and combination of plant growth regulators. The results showed that the best culture medium was MB+KT(1.0 mg/L)+NAA(0.2 mg/L), flavonoids contain in callus on the medium for 100 d reached 73.90 mg/g. accumulative dynamic state of flavonoids in callus cultured for 30 d were measured continuously, the optimal time of extracting flavonoids was determined. Flavonoids content in callus of A. venetum were increased with tissue culture technology through optimizing extraction process and screening concentration and combination of plant growth regulators.
