Determination of 6′′-O-xylosyl-tectoridin in Flos Puerariae by HPLC

Abstract: An HPLC method for determination of 6′′-O-xylosyl-tectoridin in Flos Puerariae was developed. The samples were separated on a C18 (250 mm × 4.6 mm, 5 μm) column with the mobile phase of acetonitrile-water (gradient elution). Flow rate was 0.8 mL/min. The detection wavelength was set at 265 nm. Column temperature was ambient temperature. The linear range of 6′′-O-xylosyl-tectoridin was 10.33-185.99 µg/mL, and the linear function was Y = 30731X – 216635 (r = 0.9992). The average recovery was 100.2%, and RSD was less than 2% (n = 9). The contents of 6′′-O-xylosyl-tectoridin in Flos Puerariae was 11.08-48.23 mg/g. The method was accurate and reliable, which can be used to evaluate the qualities of Flos Puerariae.
