Determination of Schisandrin in Shenan Granules by High Performance Liquid Chromatography

Abstract: The objective of this study was to develop a high performance liquid chromatography (HPLC) method for the determination of schisandrin in Shenan granules and hence to establish a quality control system of Shenan granules using liposoluble constituent as the controlling index.The chromatographic separation was performed on a Kromasil C18 column,the mobile phases consisted of methanol-water (60:40),the detection wavelength was 250 nm,flow rate was 1.0 mL/min,column temperature was room temperature,and the injection volume was 10 μL.The experimental results showed that good linear relationship for schisandrin was established between peak area and injection amount of schisandrin within the range of 0.15~6.0 μg (R2=1),the average recovery was 97.38% (RSD=2.05%).Finally,the concentration of schisandrin in Shenan granules was determined to be around 1.30 mg/g.The developed HPLC method was simple,fast,specific and reproducible.It can be used for the quality control of Shenan granules.
