Abstract: This paper reported theextraction, purification and identificationof total flavonoidsfrom root tuber of Kosteletzkya virginica. In vitro immunization and antitumor activities of the totalflavonoids were also tested. Total flavonoidsof root tuber were crude extracted by ultrasound, and were purifiedusing D101 macroporous resin. The purified compoundswereprimary identified by silica GF254 thin-layer chromatography and color reactions. The contents of total flavonoids of the crude extracts were measured by spectrophotometer. At last, biological activity ofthe totalflavonoids were tested by MTT assay. The experimental results showed that the total content of flavonoids in dry material was12.5%.The root tuber of K.virginica mainly contained flavonoids and flavonol compounds. The total flavonoidsshowed good promotion effects on thymus and spleenlymphocyteproliferation at 60 μg/mL,and showed strong inhibition effects onHepS cell proliferation at 80μg/mL.
