Identification and Determination of Polyprenols in Mulberry Leaf

Abstract: In this study,a high performance liquid chromatography (HPLC) method was developed to determine the number of isopentenyl units and the concentration of polyprenols in mulberry leaf.A Thermo C18 ODS-2 (250 mm × 4.6 mm,5 μm) column was used for chromatographic separation with methanol-isopropanol (8:7) as the mobile phases at a flow rate of 1.50 mL/min.The PDA detection wavelength was set at 210 nm,and the column oven temperature was set at 25 °C.The results showed that the number of isopentenyl units was 10 to 12 and the concentration of polyprenols in mulberry leaf was 0.731%.The developed HPLC method was accurate and reproducible.It can be used for the fast detection and determination of polyprenols in mulberry leaf.
