Effects of Albizzia julibrissin Durazz Saponins on Proliferation of Human Microvascular Endothelial Cells

Abstract: In this study,the effect of Albizzia julibrissin Durazz saponins on proliferation of microvascular endothelial cells (HMEC-1) was investigated.After 48 hour and 72hour intervention on HMEC-1 with different doses of A.julibrissin saponins,HMEC-1 proliferation and morphology were observed by Sulforhodamine B staining and optical microscope.The effects of A.julibrissin saponins on HMEC-1 were also analyzed by Trypan blue staining,DNA ladder,PI staining and flow cytometer.The results showed that A.julibrissin saponins affected HMEC-1 proliferation in time and dose-dependent manner,the IC50 of 48 hour intervention group was 1.5 μg/mL.In addition,a block was found in G0/G1 stage of mitotic HMEC-1 cycle under the saponins intervention,and cell necrosis in high dosage 48 hour intervention group.Above all,based on these results,it was concluded that cell necrosis was possibly due to cell cycle change under A.julibrissin saponins intervention,which inhibited HMEC-1 proliferation.
