Abstract: Five fractions (petroleum ether fraction,CHCl3 fraction,EtOAc fraction, n-BuOH fraction and residual fraction) obtained from 95% ethanol extract of Grifola frondosa fruiting bodies were analyzed to identify the chemical components by the system preparative tests and screened for their antitumor and DPP-IV inhibitory activities. The results indicated that compounds such as alkaloids, amino acids or proteins, organic acids, phenols, tannins, steroids and anthraquinone existed in G.frondosa fruiting bodies.Petroleum ether fraction,CHCl3 fraction,EtOAc fraction and  n-BuOH fraction showed no inhibition to WPMY-1 normal cells but showed inhibition to the proliferations of L1210,SW620,K562 tumor cells in whole or part, especially CHCl3 fraction showed strong inhibition to the proliferation of L1210 tumor cell at IC50 of 0.13 μg/mL.It indicated that these 4 fractions had antitumor activity. The residual fraction showed inhibition to L1210,K562 tumor cells and WPMY-1 normal cells, which demonstrated the residual fraction had cytotoxicity.Moreover,CHCl3 fraction,EtOAc fraction and  n-BuOH fraction showed strong inhibitory activity to DPP-IV at IC50 of 497.33,776.51 μg/mL and 704.24 μg/mL respectively. These results indicated that these 3 fractions had probability of hypoglycemic activity.
