Effects of Bound Cantharidin from Mylabris phalerata Pallas on the Apoptosis of Human Hepatoma HepG2 Cells

Abstract: Bound cantharidin was obtained from Mylabris phalerata Pallas (Maojing town of Luodian County) by water extraction method.The HepG2 cells were cultured in vitro with different concentrations of bound cantharidin.Then the annexin V-FITC/PI staining method was applied to detect the effect of bound cantharidin on apoptosis of HepG2 cells,and the expression of Bax,Bcl-2 gene in HepG2 cells were analyzed by RT-PCR.The protein expression of Bax,Bcl-2 in HepG2 cells were also detected by Western blot assay.The experimental results showed that bound cantharidin from M.phalerata-induced apoptosis of HepG2 cells,the mechanism behind may be connected with both the up-regulation expression of Bax gene and the down-regulation expression of Bcl-2 gene.
