Preparation and αAmylase Inhibition Activity of Tiliroside

Abstract: The aim of the research was to study the inhibition of tiliroside to αamylase,which was isolated from the flowers of Edgeworthia gardneri.Tiliroside was evaluated for its inhibitory activity against αamylase.The mode of inhibition for αamylase and oral sucrose tolerance test was also investigated.Tiliroside showed significant αamylase inhibitory activity (IC50 = 12.1 μM) in a noncompetitive fashion with a Ki of 9.72 μM.The results obtained from oral sucrose tolerance test revealed that tiliroside is able to inhibit αamylase at the intestinal level with consequent decrease of the postprandial peak in mice.This is the first report on the αamylase inhibition of tiliroside. In vitro and in vivo experiments showed that the inhibition of α amylase is one of the mechanisms of antihyperglycemic activity of E.gardneri.
