Effect of PGI-2 on γ-Secretase Subunit APH-1 via ERK1/2 Signaling Pathway

Abstract: In this study,the effect of COX-2 metabolic product,prostaglandin (PG) I-2 on the expression of γ-secretase subunit APH-1 was investigated via ERK1/2 signaling pathway.APH-1 expression in human neuroblastoma SH-SY5Y and mouse neuroblastoma n2a was tested by immunofluorescence.Western blot and Real-Time PCR assay demonstrated APH-1 was essential for γ-secretase activity.ELISA results showed the effect of PGI-2 on Aβ.It was found that PGI-2 treatments acceleratedthe mRNA and protein expressions of APH-1 in SH-SY5Y and n2a cells,and increased the production of Aβ.These findings provided novel insights into the mechanism of PGI2-induced AD progression.
