Abstract: In this study, the anti-tumor mechanisms of Radix Puerariae were illustrated by network pharmacology method.First, the active compounds were screened out from Radix Puerariae using drug-likeness (DL) evaluation approach.Second, the anti-tumor related protein targets were predicted by a reverse docking tool, and hand-searched in Pubmed database. Then, potential targets were submitted to MAS3.0 database and KEGG database to predict related pathway. Finally Cytoscape 3.3 software was used to construct "compounds-targets","targets-pathways" and "compounds-targets-pathways" networks. For Radix Pueraria, results showed that 16 active compounds,34 potential targets and 44 signaling pathways may participate the anti-tumor activities.Network analysis showed active compounds could affect directly on many type of cancers,or indirectly with tumor-related signaling pathway. In summary, through this study,anti-tumor mechanisms of Radix Puerariae demonstrate the features of traditional Chinese medicine (TCM) in a "multi-compounds,multi-targets and multi-pathways" mode. These provide the possible direction for the following pharmacological experiments, and gives lessons for other TCM researches.
