Optimization of Extraction of Pepsin-soluble Collagen from Antler Base Assisted by High Intensity Pulsed Electric Fields

Abstract: In order to find out the optimal extraction conditions of collagen from antler base,high intensity pulsed electric fields(PEF)-assisted pepsin methods were applied in this study.With the collagen withdrawal rate as the evaluation indicator,the effects of pepsin content,pulse number and electric field strength on extraction of collagen were studied,and the orthogonal test was designed to optimize the extraction conditions.The optimal collagen extraction conditions were determined: pepsin content 2%,pulse number 8 and electric filed strength 20 kV.cm-1,and the withdrawal rate of antler base collagen was(73.27±0.73)%.Compared with enzymatic methods,PEF-assisted extraction was less time-consuming with higher extraction efficiency.The result of SDS-PAGE confirmed that the subunits structure of pepsin-soluble collagen from antler base using PEF was mainly of α1、α2 and β.The collagen from antler base greatly maintained the characteristics of type Ⅰ collagen and with intact triple helix structure.This design can provide some reference to the extraction of collagen by PEF.
