Optimization of Extraction for Iridoid Glycosides from Lonicerae Flos by Box-Behnken Response Surface Methodology and Differences in Iridoid Glycosides Content among Different Areas

Abstract: To optimize process conditions of ultrasonic extraction of iridoid glycosides from Lonicerae Flos,with the extraction rates of loganic acid,loganin,sweroside as evaluation indexes,the single factor experiment combined with Box-Behnken design was used to optimize the extracting technology.The optimal extraction conditions were as follows:the liquid-to-solid ratio was 117 mL/g,and the ethanol concentration was 34%,the ultrasonic extraction time was 45 min,the predicted value of extraction rates was 3.28 mg/g.Under the extraction conditions after optimization,the value of extraction rates was 3.26 mg/g,and the relative deviation between the predicted and measured value was 0.31%.Meanwhile,there was great variation in iridoid glycosides content among different areas.The ultrasonic extraction technology process conditions of iridoid glycosides from Lonicerae Flos optimized by the response surface method,which was suitable and feasible.
