The studies on the fermentation optimization of Marasmius androsaceus and biological activity of its exoplysaccharide

Abstract: In order to optimize the technological conditions of liquid fermentation of Marasmius androsaceus and increase the production of crude exopolysaccharide of M. androsaceus (MEPS),the initial pH,mixing speed,fermentation temperature,inoculation amount,aeration,fermentation time,seed culture time and fermentor volume of the liquid fermentation process in 5 L fermentor were investigated.Plackett-Burman test showed thatthe significant factors were fermentation volume (X8),fermentation temperature (X3) and fermentation time (X6).Combined with the design of Box-Behnken center combination experiment and the analysis of Design-Expert.V 8.0.6.1 software,the optimum culture conditions were as follows:pH 7,rotation speed 150 rpm,inoculation volume 7%,aeration capacity 4 L/min,seed age 2 days,fermentation volume 4 L/5 L,temperature 26 ℃,time 5.6 days.Under the optimized processes,the response value of polysaccharide yield was 204.27 mg/g.The mice weight-bearing swimming test and endurance running experiment showed that the MEPS has good endurance and anti-fatigue effects.
