Protective effects of Chinese hawthorn leaf hyperoside against high glucose-induced injury in SH-SY5Y cells

Abstract: This study was to explore the protective effect and mechanisms of hyperoside(Hyp) against oxidative stress of SH-SY5Y cells induced by high glucose.SH-SY5Y cells in 100 mmol/L glucose medium was incubated with 20,50,100 μmoL/L hyperoside for 36 h.The cell proliferation of SH-SY5Y,the levels of lactate dehydrogenase (LDH),intracellular reactive oxygen species (ROS) as well as the contents of malondialdehyde(MDA) and glutathione(GSH),activities of caspase-3,superoxide dismutase (SOD) and catalase (CAT) were measured.Both mRNA expression levels and protein expression levels of NF-кB and SIRT1 genes were detected.The treatment of SH-SY5Y cells with hyperoside had a marked diminution in LDH level,caspase-3 activity,intracellular levels of MDA and ROS.Whereas,the cell viability,the activities of SOD and CAT,GSH contents,the mRNA expressions and the protein contents of SIRT1 were notably elevated and the mRNA and the protein expression of NF-кB were attenuated by hyperoside.The results indicated that hyperoside may protect against the oxidative damage of SH-SY5Y cells induced by high glucose via the activation of SIRT1 and inhibition NF-кB.
