Study on the relationship between antioxidant activity of Rehmannia root and spectral effect based on HPLC fingerprint technology

Abstract: In this study,the antioxidant activity in different tissues and different polar parts of R. glutinosa was compared and preliminary analysis of its antioxidant activity spectrum relationship.We evaluted the antioxidant activity in different polar position(chloroform, n-butanol and ethyl acetate) of radial striation(xylem),unradial striation(phloem) and whole roots of two R. glutinosa germplasms(855,Beijing1) by the method of DPPH.And using Pearson correlation coefficient method to analyze the spectral effect relationship between HPLC fingerprint and antioxidant activity of strong antioxidant activity fraction.The results showed that the radial striation,unradial striation and whole root tuber of Rehmannia glutinosa root had strong antioxidant activity,in which the overall antioxidant capacity of the radial striation was weak,and the most active was the ethyl acetate part.The results of oneway ANOVA showed significant differences in IC50 values between ethyl acetate fraction and the antioxidant activities of chloroform and n-butanol fractions.HPLC fingerprints of the ethyl acetate fraction of the sample were calibrated with 14 common peaks.The results of spectral efficiency analysis showed that the acteoside had certain antioxidant activity,and the peaks X1,X 2, X10 and ethyl acetate samples had significant correlation with oxidation resistance.This study provides a theoretical basis for the screening of antioxidative activity pharmacological components of Rehmannia glutinosa and the clarification of the relationship between the radial striation of Rehmannia glutinosa and the quality of Rehmannia glutinosa.
