Abstract: To prepare water extract of Eucommia ulmoides Oliv. from Guangyuan and commercial Eucommia ulmoides  Oliv.(EUO) of different grades,in order to compare the effects on enhancing immune function and protecting liver function among them.Through the immunosuppressive mice models which were made by injecting hydrocortisone subcutaneously,the immunity-enhancement capability of three kinds of EUO was tested by detecting the carbon clearance index K,the phagocytic index α,and the index of immune organs.Through the immunosuppressive mice models which were made by injecting cyclophosphamide subcutaneously,the immunity-enhancement capability of three kinds of EUO was tested by detecting the number of peripheral blood cells and the index of immune organs.The acute liver injury model was induced by injecting CCl₄ into mice abdominal cavity,then judge the effect of three kinds of EUO on liver protection by detecting the liver index,the levels of ALT and AST in serum,the SOD and MDA levels in liver tissues.As the result,the water extract of Guangyuan EUO could increase the phagocytic index α of immunosuppressive mice greatly (P<0.01),all water extract of EUO could make PLT counts of immunosuppressive mice tend to be normal,while the second-class EUO obviously increased the RBC counts (P<0.05).Comparing with acute liver injury mice,all drug-treated group had a significant effect on decreasing the levels of ALT and AST in serum,and Guangyuan EUO reduced the ALT level evidently (P<0.01),while the AST level was distinctly lowered by second-class EUO (P<0.01),third-class EUO and Guangyuan EUO (P<0.05).All drug treated groups obviously improved the level of SOD in liver tissues (P<0.01),and decreased MDA level in liver tissues evidently.Second-class and third-class EUO can lower the level of MDA in liver tissues distinctly (P<0.01).In conclusion,EUO from Guangyuan and commercial EUO of different grades all have effects on immunity-enhancement and liver protection.EUO from Guangyuan has the most obvious effect on immunity enhancement,while the second-class EUO’s effect on liver protection was the best.
