Study on isobutyl amide compounds from Zanthoxylum bungeanum and their activities against ferroptosis in HT22 hippocampal cells

Abstract: The chemical constituents of anthoxylum bungeanum collected in Luding County of Sichuan Province were investigated in order to develop new lead compound.Eight isobutyl amide compounds were isolated and identified from 70% hydrochloric acid extract of Z. bungeanum by silica gel column chromatography and high performance liquid chromatography.They were elucidated as zanthoxylumol A(1),hydroxy-α-sanshool(2),tetrahydrobungeanool(3),hydroxy-γ-isosanshool(4),hydroxy-γ-sanshool(5),bungeanool(6), γ-sanshool(7),isobungeol(8),and β-sitosterol(9) by high resolution mass spectrometry and nuclear magnetic resonance spectroscopy.Compound 1 was isolated from this plant for the first time.Compound 3 demonstrated good inhibitory effect on the hippocampal neuron cell line HT22 erastin-induced ferroptosis with IC50 to be 4.08 μM.
