Effect of taurine on triglyceride synthesis in HepG2 cells

Abstract:

The effect of taurine on triglyceride synthesis and its preliminary mechanism in oleic acid-treated HepG2 cells was researched in this article.A high triglyceride cell model was established by adding 0.05 mmol/L oleic acid to DMEM medium.HepG2 cells were treated with taurine at final concentration of 1,5,10 and 20 mmol/L for 24,48 and 72 h,and the levels of intracellular triglycerides were determined.The expression of SREBP-1c and the enzymes of AceCS1,ACC,FAS and ACSL1,associated with fatty acid synthesis,were detected by western blot after HepG2 cells were incubated with 5 mmol/L taurine for 24 h.Intracellular triglycerides levels were decreased by 1 mmol/L taurine treatment for 72 h (P<0.05),5 and 10 mmol/L taurine treatment for 24/48/72 h (P<0.05) and 20 mmol/L taurine treatment for 24/48 h (P<0.05) in high-triglyceride cells.And the protein expression of SREBP-1c,FAS,ACC,AceCS1 and ACSL1 were reduced while p-ACC was increased by 5 mmol/L taurine treatment for 24 h (P<0.05).Taurine inhibits the synthesis of fatty acid/triglyceride in oleic acid-treated HepG2 cells by regulating SREBP-1c and its downstream genes.
