Effects of vitexin on antioxidant enzyme activity and related gene expression in type 1 diabetic mice

Abstract:

To investigate the antioxidant effect of vitexin on type 1 diabetic mice,84 four-week-old SPF-grade male ICR mice were randomly divided into different control groups based on body weight of mice in this study,including blank control group,water model control group,DMSO model control group,model groups with high-,medium- and low-dose of vitexin,and positive control group.The difference in body weight of mice among control groups was not significant.After successful modelling and continuous gavage for mice with different doses of vitexin for 4 weeks,enzyme activities (SOD,GSH-PX and T-AOC) and MDA content in serum and liver tissues of mice were measured by the reagent kit method;the expression of antioxidant genes SOD-1,SOD-2,GPX-1 andGPX-4 were measured by quantitative fluorescence PCR.The results have shown that vitexin can highly significantly increase enzyme activities (SOD,T-AOC and GSH-PX),and highly significantly decrease MDA content,and significantly up-regulate gene expressions of SOD-1,SOD-2,GPX-1 andGPX-4  in type 1 diabetic mice.Therefore,vitexin can improve the antioxidant capacity of type 1 diabetic mice.
