Optimization of Decolorization Technology of Cornus officinalis Sieb.et Zucc Polysaccharide by Macroporous Resin

Abstract: The resin with better decolorization effect was screened out from D301,D303,D315 and D900 resin using decolorization rate,polysaccharide retention rate and cell inhibition rate as evaluation indexes.The inhibition effects of Cornus officinalis Sieb.et Zucc polysaccharide before and after decolorization on HL-60 cells were investigated by MTT assay.D303 resin showed the best decolorization effect.Its processing conditions were further optimized by single factor and orthogonal experiments.The results showed that the optimal process condition were as follows:processing time of 7 h,temperature of 50 ℃,sample concentration of 15 mg/mL.Under the optimized conditions,the decolorization rate and polysaccharide retention rate were 79.71% and 81.12%,respectively.The developed decolorization technology using macroporous resin for C.officinalis polysaccharide provided experimental basis for its further research and development.
