Abstract: PTP1B plays an important role in the negative regulation of insulin signaling pathway.Overexpression of it leads to dephosphorylate the tyrosine residues of insulin receptor proteins, which is primarily responsible for insulin resistance in type 2 diabetes.Therefore,PTP1B are potential target for screening active ingredients of diabetes treatment.Inhibitory of PTP1B oriented, two specific Uygur herbs Eremurus altaicus and Eremurus anisopterus showed good inhibitory effect on PTP1B with IC50 of 17.85 and 33.76 μg/mL.The compound isolated from E.altaicus showed good inhibitory effect with IC50 of 26.15± 0.5 μM; The enzyme kinetic studies revealed mixed-type inhibition type with the inhibitory constants (Ki) of 39.12±8.56 μM.The study provided active guidance and theoretical basis for further isolation and purification of E.anisopterus.
