Optimization of Micro Screening Model for Acetyl Cholinesterase Inhibitors

Abstract: To optimize the micro screening assay for acetylcholinesterase inhibitors(AChEIs),wavelength,incubation time,substrate concentration,enzyme concentration and the solvent used in the screening were evaluated.Physostigmine and huperzine A were tested as positive drugs to verify the reliability of the model.Results showed that the micro screening model for AChEIs was optimized with wavelength 410 nm,incubation time 20 min,substrate concentration 4 mmol/L,AChE concentration 0.5U/mL.In addition,50% aqueous methanol was selected as an unified solvent to dissolve the samples in the screening.Physostigmine and huperzine A test indicated that the optimized micro screening system is stable,reliable and sensitive that can be successfully used to largescale screening of AChEIs.
