Inhibition Effects of Rodgersia aesculifolia Batal Extracts on Human Gastric Cancer Cells MGC80-3

Abstract: Human gastric cancer cells were cultured with different concentrations of ethyl acetate extract and ethanol extract from Rodgersia aesculifolia Batal.MTT method was used to observe the effects of the extracts on gastric cancer cell proliferation.Immunocytochemistry was used to detect the Bcl-2 and Bax expression of gastric cancer cells.Using ELISA method to detect PGE-2 production,so that to explore the inhibition mechanism of R.aesculifolia extracts on gastric cancer cell.Results showed that different concentrations of two kinds of extract can inhibit gastric cancer cell proliferation,inhibition was significant after 48 hours and 72 hours.Extracts with concentrations of 40 mg/mL and 10 mg/mL,can reduce the Bcl-2 expression and enhance the expression of Bax significantly (P<0.05).The extracts with all the concentration levels can reduce the generation of PGE-2 of gastric cancer cells.Hence,it was concluded that R.aesculifolia extracts promoted cell apoptosis possibly by adjusting the expression of Bcl-2 and Bax and inhibited the proliferation and invasion of gastric cancer cells possibly by reducing the generation of PGE-2.
