Screening of Anti-diabetic Active Fractions from South Schisandra with Vinegar in vivo and in vitro

Abstract: In this study,the antidiabetic active fraction of Schisandra chinensis was screened in vitro and in vivo using the inhibition rates of α-amylase and α-glucosidase as evaluation index.Mice were intraperitoneally injected with streptozotocin(STZ,140 mg/kg) to develop diabetic animal model;Alanine aminotransferase(GPT),high density lipoprotein cholesterol(HDL-C),total cholesterol(T-CHO),total triglycerides(TG)in the serum,malondialdehyde(MDA) and superoxide dismutase(SOD) in the liver tissue were measured.The effects of four chemical fractions(Petroleum ether,chloroform,n-butanol,water)were difference.The chemical fraction of petroleum ether from S.chinensis showed the best inhibition effect on α-amylase;Petroleum ether and chloroform fractions showed the best inhibition effect on α-glucosidase.The in vivo study showed that each administration group can lower blood sugar,improve insulin content,and enhance the mice glucose tolerance,but the effect of petroleum ether and chloroform fractions was obvious.The effects on HDL-C,GOT,GPT,TG and T-CHO were significantly different in the petroleum ether and chloroform fractions.The effect on MDA and SOD contents in the liver tissue of petroleum ether and chloroform fractions were obvious,but there was no obvious change in the other fractions.Both results of in vitro and in vivo pharmacodynamic evaluation showed that the antidiabetic effect of petroleum ether and chloroform chemical fractionswas obvious.
