Effect of Matrine and Oxymatrine on Proliferation and Energy Metabolism of ECV304

Abstract: The aim of this study was to investigate the effect of matrine and oxymatrine combination on the activity and energy metabolism of human umbilical vein endothelial cells(ECV304).The method of permutation and combination was used to determine the best ratio of matrine and oxymatrine concentration.The cell growth curves were used to describe the effect of matrine and oxymatrine combination on cell proliferation.The morphological changes of ECV304 cells were determined by HE staining and transmission electron microscopy.The enzymatic activity and metabolite quantity of the metabolite related enzymes in ECV304 cells was also measured.Results showed that matrine at 1200 μg/mL and oxymatrine at 600 μg/mL combination had a better effect.The proliferation of ECV304 cells was significantly inhibited by the combined usage of matrine and oxymatrine and the cells morphology were also changed significantly as compared with control group and matrine/oxymatrine group.The combination usage of these two drugs did not affect HK activity,PK activity and lactate metabolism in glycolysis,but can significantly reduce SDH activity and ATP level in oxidation process.Combined usage of matrine and oxymatrine had an indirectly antitumor effect in ECV304 cells by inhibiting cell proliferation and tumor angiogenesis.
