Analysis of Chemical Constituents in Polygoni Multiflori Radix and Polygoni Multifori Caulis from the Same Origin Based on UPLC-Triple TOF-MS/MS

Abstract: The difference of chemical composition in Polygoni Multiflori Radix (PMR) and Polygoni Multifori Caulis (PMC) from the same origin was analyzed by ultra-performance liquid chromatography coupled with triple quadrupole-time of flight tandem mass spectrometry (UPLC-Triple TOF-MS/MS).The samples were extracted with methanol by ultrasonication at room temperature and centrifuged.Through the analysis of the multistage tandem mass spectrometry,the characteristic peaks were extracted with mass spectrometry data peak matching,peak alignment,noise filtering.The obtained data were analyzed by principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA).The components were identified according to MS accurate mass and MS/MS spectrometry fragmentation information,combined with the software of database searching and literature component identification.The results showed that the chemical compositions between PMR and PMC from the same origin were clearly distinguished and 42 chemical compositions were identified.Among of them,12 compositions were common chemical composition presented in different changing laws.The research provided experimental data for revealing the laws on metabolites of PMR and PMC from the same origin.
