Separation,Purification and Antibacterial Activity of Iridoid glycosides from Gentiana macrophylla Pall.

Abstract: The aim of this study was to optimize the separation and purification technology of iridoid glycosides from Gentiana macrophylla Pall.with XDA-1 macroporous adsorption resin.The antibacterial activity of the purified iridoid glycosides were studied in vitro.With the content of gentiopicroside as index,dynamic adsorption-separation was used to determine the optimal separation and purification conditions of iridoid glycoside from G.macrophylla.The optimal conditions of XDA-1 macroporous adsorption resin were finally determined as follows:sample injection concentration was 0.07 g original/mL,pH was 5.0,sample flow rate was 4 BV/h,quantity of adsorption was 32 BV,desorption rate was 3 BV/h,the pH was 7.0 and elution volume was 10 BV of 50% ethanol.The purity of obtained iridoid glycosides was 62.97%.Moreover,the antibacterial activity of the extract was evaluated by 3 bacteria inhibition zone and minimum inhibitory concentration.The results showed that the obtained iridoid glycosides had good antibacterial activity.
