Optimization of Ethanol Reflux Extraction of Phenylethanoid Glycosides from the Leaves of Callicarpa nudiflora by Response Surface Methodology

Abstract: The study was aimed to optimize the ethanol reflux extraction conditions of phenylethanoid glycosides (PhGs) from the leaves of Callicarpa nudiflora by response surface methodology with the extraction yield of PhGs as index.Six factors such as particle size,soaking time,extraction duration,times of extraction,ethanol concentration and solvent-material ratio were investigated on the basis of single-factor experiments.The results showed that the optimal ethanol extraction conditions were optimized as follows:extraction duration 120 min,ethanol concentration 89% and 20 times of solvent-material ratio.Under these conditions,the mean extraction yield of PhGs was 11.83%,whose deviation was small by comparing with the predicted model value 11.86%.The result proved that the optimized extraction process was stable and reliable,and the optimized parameters can provide guidance on the industrialization of ethanol reflux extraction of it.
