Protective Effect of Mangiferin on Alcoholic Hepatitis in Rats

Abstract: To study the protective effect and mechanism of mangiferin (MFN) on alcoholic hepatitis (AH) in rats.SD rats were randomly divided into normal group,model group,silibinin group (SYN,20 mg/kg) and MFN high-dose,low-dose groups (100,50 mg/kg).An AH rat model was established by intragastric administration of 40% ethanol (10 mL/kg),medication was given at the same time.After 4 weeks of drug delivery,the concentrations of serum ALT and AST,TBIL,DBIL and IBIL were detected.The mRNA level of NF-κB and the contents of TNF-α and IL-6 in liver tissues were measured.Hematoxylin eosin (HE) staining and Oil red O staining were used to observe the pathological changes of liver tissue.Compared with normal group,the serum indexes of ALT,AST,TBIL,DBIL and IBIL were increased (P<0.01).NF-κB mRNA ,TNF-α and IL-6 levels in liver tissue were increased (P<0.01).At the same time,a large number of vacuolar lesions,swelling of liver cells,fatty degeneration and inflammatory cell infiltration were observed in the liver.Compared with model group,the above-mentioned indexes in MFN high and low dose groups were significantly improved (P<0.01, P<0.05),and the degree of liver cell lesion was obviously alleviated and dose dependent.Taken together,the result suggested that MFN has a good anti-AH effects,the mechanism of action is related to hepatocyte repair,regulation of liver cell inflammation (especially NF-κB,TNF-α,IL-6 levels).
