The Proliferation Effects of 3,3′-dimethyl ellagic acid-4-xylycoside, An Active Component from Sanguisorba officinalis L.on The NIH3T3 Cell and Its Associated Mechanism

Abstract: In present study,we aimed to investigate 3,3′-dimethyl ellagic acid-4-xylycoside (DEAX) of Sanguisorba officinalis L.whether it can promote the NIH3T3 cells proliferation & migration,also to explore its associated mechanism.The purity analysis and structure identification of DEAX were performed by Agilent 6230 UPLC-DAD-TOF-MS.The proliferations on NIH3T3 cells treated with the different DEAX concentrations and time points were detected by MTS assay,and the migrations of NIH3T3 cells treated with different DEAX concentrations were determined by scratch assay.The concentrations of hydroxyproline (HYP) and transforming grow factor-β1 (TGF-β1),and the mRNA expressions of TGF-β1and Collagen I were measured by ELISA and qRT-PCR,respectively.The results showed that DEAX could significantly promote the proliferation and migration of NIH3T3 cell (P<0.05 or P<0.01),and improve the protein and mRNA expressions of TGF-β1 and Collagen I (P<0.05 or P<0.01).These results indicated that DEAX could promote the proliferation and migration of NIH3T3 cell through improving the protein and mRNA expression of TGF-β1 and Collagen I,which provided scientific evidence for the material basis and mechanism of wound healing in Sanguisorba officinalis L.
