Rapid characterization of triterpenoid saponins in Achyranthes bidentate Blume by UPLC-Q-TOF/MS in MSE mode combined with UNIFI screening platform

Abstract: To investigate the main chemical constituents of Achyranthes bidentate triterpenoid saponins,a rapid qualitative analysis of triterpenoid saponins in Achyranthes bidentata was carried out by ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) in MSE mode combined with UNIFI scientific information system.Firstly,the chemical constituents of Achyranthes bidentata triterpenoid saponins were separated by UPLC chromatography and the mass spectrometry data were collected by QTOF/MS in MSE mode.Then,the chemical constituents of Achyranthes bidentata triterpenoid saponins were screened automatically through UNIFI platform and checked by MassLynx workstation,so as to quickly complete the qualitative analysis of the chemical constituents of Achyranthes bidentata triterpenoid saponins.AS a result,forty triterpenoid saponins were isolated and identified in this experiment.Among them,there are 38 kinds of compounds with oleanolic acid as the mother nucleus,and two triterpenoid saponins with other mother nucleus.This method can quickly analyze the triterpenoid saponins in Achyranthes bidentata,which will provide reference for the quality control of Achyranthes bidentata saponins and the study of subsequent pharmacodynamic substances.
