Optimization of extraction process of angelica by orthogonal test with three active ingredient targets

Abstract: Orthogonal test was applied to optimize the extraction process of angelica which was extracted by alcohol first and water later,with ligustilide,ferulic acid and polysaccharide as evaluation indexes.The effects of extraction frequency,concentration of ethanol,extraction temperature,solid-liquid ratio and extraction time on ethanol extraction technology were investigated by using orthogonal table L16(45) test with ligustilide and ferulic acid as indexes.Optimize ethanol extraction technology was extraction frequency (twice),concentration of ethanol (80%),extraction temperature (85 ℃),solid-liquid ratio (8∶1) and extraction time (2 hours for each time).The effects of extraction frequency,extraction temperature,solid-liquid ratio and extraction time on water extraction technology were investigated by using orthogonal table L9(34) test with polysaccharide as index.Optimize water extraction technology was extraction frequency (three times),extraction temperature (100 ℃),solid-liquid ratio (9∶1) and extraction time (1 hour for each time).The optimum ethanol and water extraction process showed good repeatability,stability and feasibility.
