Study on enzymatic extraction process of Terminalia chebula Retz and Rubia cordifolia L. processed Terminalia chebula Retz polysaccharide

Abstract: To optimize cellulase-assisted extraction process of Terminalia chebula Retz and Rubia cordifolia L. processed Terminalia chebula Retz polysaccharide.This experiment taking the Terminalia chebula Retz and Rubia cordifolia L. processed Terminalia chebula Retz as the research object,the extraction rate of polysaccharides is an index.The effects of ratio of material to liquid,extraction temperature,extraction times and enzym concentration on the extraction rate of polysaccharides from Terminalia chebula Retz and Rubia cordifolia L. processed Terminalia chebula Retz were studied by single factor experiment and orthogonal experiment,screening for optimal extraction conditions.The best extraction process for extracting water-soluble crude polysaccharides from Terminalia chebula Retz by the cellulase method was determined as follows:solid-liquid ratio is 1∶50,extraction temperature is 50 ℃,extraction time is 60 min and the enzyme dosage is 1.8%.Under these conditions,the maximum extraction rate of polysaccharide was 2.92%.The best extraction process for extracting water-soluble crude polysaccharides from Rubia cordifolia L. processed Terminalia chebula Retz by the cellulase method was determined as follows∶solid-liquid ratio is 1∶40,extraction temperature is 40 ℃,extraction time is 90 min and the enzyme dosage is 1.8%.Under these conditions,the maximum extraction rate of polysaccharide was 3.29%.The extraction rate of polysaccharides from Rubia cordifolia L. processed Terminalia chebula Retz was better than from Terminalia chebula Retz.The process is low energy consumption,safe,effective and less damage to polysaccharides.It can lay a theoretical foundation for industrialized production of polysaccharides extracted from Terminalia chebula Retz and Rubia cordifolia L. processed Terminalia chebula Retz.
