Screening of Bacillus tequilensis WRN032 with Magnaporthe oryzae and its optimization of fermentation conditions

Abstract: The Oxford cup method was used for bacteriostasis test,and the growth inhibition effects of different phytopathogenic fungi were screened according to a variety of Bacillus species,and Bacillus tequilensis WRN032 had the best inhibitory effect on the growth of Magnaporthe oryzae.Using the absorbance and the inhibition zone diameter as indicators,the single-factor test,Plackeet-Burmann test and Central Composite test were designed to optimize the fermentation conditions of Bacillus tequilensis WRN032.The optimal fermentation conditions for the antibacterial activity of the strain were:inoculation volume 1%,flask filling volume 40%,glucose concentration 5%,shaker speed 220 rpm,initial pH6.5,incubation time 48 h,incubation temperature 30 °C.Under these conditions,the absorbance of the fermentation broth and the inhibition zone diameter of Bacillus tequilensis WRN032 increased by 186.63% and 154.10%,respectively,compared with initial fermentation.
