Effect of jujube leaves alcohol extract on M1 and M2 polarization of mouse macrophages

Abstract:

To explore the effect of jujube leaves alcohol extract (JLE) on M1 and M2 polarization of RAW264.7 cells.In this study,RAW264.7 cells were polarized into M1 and M2 macrophages by LPS or IL-4,respectively.CCK8 was used to detect the cell survival rate.Griess method was used to detect the effect of drugs on the release of NO in M1 macrophages.RT-PCR was used to detect the expression of M1 and M2 marker genes.Scratch test and RT-PCR were used to detect the migration of 4T1 cells treated by M2 macrophages culture supernatant.The results showed that the jujube leaves alcohol extract could inhibit the release of NO,down-regulate IL-12p40,IL-1β and COX-2 in M1 macropage and Arg1,CD206 in M2 macrophage.Further,it could reduce the mRNA level of MMP-2,MMP-9 and NF-κBp65 in 4T1 under the influence of M2 macrophages,and inhibit the migration of 4T1 cells.In conclusion,the jujube leaves alcohol extract can inhibit M1/M2 polarization of macrophages,and inhibit the migration of 4T1 cells through NF-κB pathway.
