The network pharmacology study of Strychnos nux-vomica L. in treating rheumatoid arthritis

Abstract:

To screen the active ingredients and potential mechanism of Strychnos nux-vomica L. in the treatment of rheumatoid arthritis by Chinese medicine network pharmacology.The TCMSP database was used to select the active ingredients and targets of S. nux-vomica,and then the Gene name annotation was performed using the UniProt database,and the disease-related targets were retrieved in the GeneCards database.The Rversion 3.6.2 software and Cytoscape 3.7.1 software was used to predict the targets and active components of S. nux-vomica. for RA treatment,and the interactions between proteins were evaluated using the STRING database.Finally,the BiocManager package in R language was used for GO analysis and KEGG pathway enrichment analysis.A total of 13 effective components of S. nux-vomica and 23 “Traditional Chinese medical” -“disease” targets were screened through the above process,and the core targets involved PTGS2,NR3C1,ESR1,MAOA,OPRM1,SLC6A4,etc.GO function enrichment analysis got 123 GO entries,and KEGG pathway enriched to obtain 34 signal pathways closely related to the onset of rheumatoid arthritis,such as interleukin (IL) - 17 signaling pathways,tumor necrosis factor (TNF) signaling pathway,the NF-κ B signaling pathways,toll-like receptors signaling pathways,apoptosis pathway,hypoxia inducing factor (HIF)-1 signaling pathways,T cell signaling pathways,B cell signaling pathways,MAPK signaling pathways,VEGF signaling pathways,JAK-STAT signal pathway and osteoclast differentiation pathway.This study predicts the material basis and potential mechanism of S.nux-vomica in the treatment of rheumatoid arthritis,and provides a theoretical basis for the further development of S.nux-vomica.
