Study on the protective effect and mechanism of rape bee pollen ethanol extract on cardiomyocyte hypertrophy induced by isoproterenol

Abstract:

In order to investigate the protective effect and mechanism of rape bee pollen ethanol extract (RBPEE) on isoproterenol (ISO)-induced H9c2 cardiomyocyte hypertrophy,in this experiment,the cardiomyocytes were treated with gradient concentration of RBPEE for 24 h,and starved for 12 h,and then injured with 50 μmol/L ISO for 48 h,and the treated cells were collected for cytoskeleton staining,surface area,total protein,SOD,GSH and MDA determination,and related gene expression level detection.The study showed that RBPEE could significantly reduce the surface area,total protein content and the mRNA expression levels of cardiomyocyte hypertrophy markers ANP,BNP and β-MHC of the hypertrophic cardiomyocytes,and significantly increase SOD activity and GSH content,and decrease MDA content,as well as significantly reduce the mRNA expression levels of inflammatory response-related factors IL-2,IL-6,TNF-α and apoptosis-related factors Caspase-3,Caspase-8,Caspase-9 and Bax,and significantly increase Bcl-2 mRNA expression level and Bcl-2/Bax.The results suggested that the protective effect of RBPEE on ISO induced cardiomyocyte hypertrophy was closely related to the reduction of oxidative stress level,the inhibition of inflammatory response and the anti-apoptotic effect.
